Abstract:

Introduction:Boosting immune system of human body can be a way in control and treatment of cancer. Colorectal cancer is one of the reported common cancers whichshow highly incidence rate of death. Applying nanoparticles as a gene delivery system can be effective in manipulation of gene expression. Materials and methods: IL-4R was targeted to be more expressed through multiplying of vector plasmid containing human IL-4 receptor gene DNA. Transformation of plasmid DNA-hIL-4R into competent cells of E.coli XL-1 blue was done to make recombinant DNA clone. DNA of transformed cells was purified using Wizard plus SV Miniprep. Agarose gel electrophoresis was run to confirm presence of recombinant DNA. Magnetic nanoparticles were used in transient transfection of HT-29 colorectal cancer cells using magnetofection technology. Results:The percentage of dead cells was found through cell proliferation assay. The obtained data was analyzed by using Student’s t-test. The results indicated that upregulation of IL-4R gene caused its internalization and activation after binding to IL- 4 in cancer cells. Conclusion: There is a possibility of tumor regression after transfection of HT-29 tumor cells with IL-4R due to over-expression of STAT6 which mimic IL-4 function.
