Ovary and hypothalamic adiponectin geneexpressions in PCOS model rats following Ldopa injection

Abstract:

Adiponectin hormone is synthesized in the hypothalamus, ovary and adipose tissue. Plasma adiponectin levels and dopaminergic neurons activity is low in polycystic ovary syndrome (PCOS). Dopamine exerts stimulatory effects on adiponectin secretion in intact individuals. In the present study the effects of L-dopa was investigated on relative adiponectin gene expressions in the hypothalamus and ovary of PCOS model rats. PCOS condition was induced in fifteen Wistar female rats weighing 180-220g by intramuscular injections of estradiol valerate. The PCOS rats in two groups received saline or L-dopa (100mg/kg) via intraperitoneal injection respectively. Five intact estrous rats as a control group received saline intraperitoneally. Hypothalamic and ovary samples were dissected. Mean relative adiponectin gene expressions were determined by real time polymerase chain reaction (RT-PCR) method. Hypothalamic and ovary adiponectin gene expressions significantly decreased in PCOS rats in comparison to intact ones. L-dopa significantly increased hypothalamic adiponectin gene expressions in comparison to PCOS rats. But injections of L-dopa did not significantly increase the ovary adiponectin geneexpressions compared to L-dopa group. Dopaminergic neurons may be involved in the controlling the neuroendocrine axis of PCOS via regulating hypothalamic adiponectin synthesis.
