Abstract:

In this study, ultrasound - assisted ionic liquid based dispersive liquid - liquid microextraction method (UA-IL-DLLME) coupled with high performance liquid chromatography (HPLC) administered for extraction and determination of two antidepressant drugs dosages including fluvoxamine and sertraline in human plasma and urine samples. Different factors could affect the extraction steps and efficiency of extraction. Sample solution pH, the type and volume of extraction solvent, the time of ultrasonic, the time and rate of centrifuge rotation and ionic solution strength are some factors that affect UA-IL-DLLME method. In this research, optimum conditions were gained by: pH = 9, [C8MIM] [PF6] ionic liquid, 30 μl of ionic liquid, 5 min ultrasonic time, 5 min centrifugation time, centrifuge rotation rate of 4000 rpm, 2 min extraction time and no salt increase. In quantitative experiments, limit of detection (LOD) of 0.2 μg/L for fluvoxamine and 0.7 μg/L for sertraline and preconcentration factor (PF) of 1379 for fluvoxamine and 1156 for sertraline was determined. Obtained regression coefficients (r2) were 0.999 for fluvoxamine and sertraline. Finally UA-IL-DLLME coupled with HPLC was successfully used for detection of fluvoxamine and sertraline in human plasma and urine samples.
