Abstract:

Introduction & Aim: After lung cancer colorectal cancer is the second most common cancer. cucurbitacins are natural compounds previously shown to have antitumor activities as well as a potent chemo-preventive action against cancer. Methods: In this research, first, using the colorimetric MTT test, the LC50 was obtained. to study the changes of genes expression, cellular RNA was extracted and studied using Real-time RT-PCR method after cDNA synthesis. Results: The expression of BAX was suppressed in all treatments of cucurbitacin E, D and I whereas the level expression of Caspase-3 had a significant increase in all three types of cucurbitacin. The p53 gene expression in cucurbitacine D was extremely different from cucurbinatin E and I so that it had a larger number and the highest level of expression for the p53 gene. Also the increase in level of AIF and BCL2 was observed. cucu I treatments had a larger number Although the level of gene expression was partly similar. Conclusion: As other studies and our previous MTT results have been shown cucurbitacins are cytotoxic agents against cancer cell lines. Our apoptosis gene expressions paradigm in Real-Time results show a non-classical apoptosis pathway. There is a number of researches that report contradictory gene expression result compare to classical apoptosis pathway.
