Abstract:Lung cancer is the leading cause of death worldwide. MicroRNAs (miRNAs) are endogenously processed, single-stranded, small (16-26nt) non-coding RNAs which post-transcriptionally regulate theexpression of most protein-coding genes in human. By binding to and suppressing the translation oftheir target mRNAs, miRNAs control important cellular processes such as proliferation, differentiation, apoptosis, and tumor-genesis. A recent study showed that most of the miRNA genes are located at fragile and cancer related chromosomal regions. Now, it is estimated that the number could reach50,000 miRNAs all over the genome. We found a frequently amplified genomic region in lung cancer, which are very potential to hold functional miRNAs (oncomirs) that suppress certain tumor suppressor genes. Based on the available cytogenetic data, 7p, is a frequently amplified region in lung cancers, wasscanned for novel miRNAs. Using the SSC profiler, MiPred, mireVal, MatureBayes, CID-miRNAsoftwares, the potential regions of the 7p11-13 sub-band were scanned for finding stem-loop structures.Finally 2 stem-loop structures were selected for further experimental verification. Then specific primerswere designed in order to amplify and clone the candidate sequences containing the stem-loopstructure. After that the amplified sequence will be cloned into an expression vector in order to be transfected in human cell lines. The clones will be over-expressed in cell-lines to verify that they generate a mature miRNA from a pre-mir precursor. These novel miRNAs which seem to be functional in lung cancer have a potential to be used as diagnostic biomarkers for early detection of lung cancer.
