Abstract:Multiple sclerosis is a chronic inflammatory disease of the central nervous system. The main cause of MS is unclear. Some changes of protein expression such as apolipoproteinE in MS patients have been confirmed in previous studies. ApoE is a 34.2 KDa glycoprotein and contains 229 amino acids. Homeostasis maintenance of cholesterol in plasma and central nervous system is the main function of ApoE. The nonlipid-related properties of apoE include modulating inflammation andoxidation, suppressing T cell proliferation, regulating macrophage functions, and facilitating lipid antigen presentation byCD1 molecules to natural killer T cells (NKT). Transcriptional profils of PBMC in Blood can be used to identify theireffects in the MS disease. Therefore, in this study, we have analyzed the effects of the inflammatory process in the central nervous system on the transcription of the ApoE gene in peripheral blood mononuclear cell in quit race MS patients and healthy controls of the North West of Iran. Thus, we attempt to evaluate the expression level of ApoE in PBMC of MSpatients in comparison with normal individuals by Real-time PCR. The PBMC was separated from the whole blood of 45patients and 30 normal cases by Ficoll-hypaque. The total cellular RNA was extracted and then cDNA was synthesized from total mRNA of samples. This process was followed by Real-time PCR using primer pairs specific for ApoE mRNA and beta-actin as internal control. Results revealed that there were no significant expression changes in groups of patients compared to the control group
