Abstract:
Background and aim: Autophagy, a highly conserved self-digestion process, is one of the mechanisms that may play a dual role in the development or inhibition of cancer. Different signaling pathways have been involved in the up regulation or down regulation of autophagy. MicroRNAs are small noncoding RNAs that have important roles in regulation of gene expression. This study evaluated the expression levels of mir-133 and mir-199 and potential target genes expression of autophagy pathway such as FIP200 and ATG13 in breast cancer patients. Methods: this case-control study was performed in 47 cancerous tissue samples and pair-matched adjacent non-cancerous tissues were obtained from breast cancer patients. The samples were collected with ethical principles of Imam Khomeini hospital cancer institute using American Joint Committee on Cancer (AJCC) guideline. After RNA extraction, cDNA synthesis and qRT-PCR, the expression levels of FIP200, ATG13 and mir-133, mir-199 was measured and results were confirmed by western blot.Result: our results have shown that the expression levels of FIP and ATG13 decreased in tumor samples versus non-tumor samples. Also, irregulation of mir-133, mir-199 expression have been shown by qRT-PCR.Conclusion: We suggested that mir-133, mir-199 play pivotal roles in regulation of FIP200 and ATG13 that involve in autophagy process. Accordingly, it can be said that mir-133, mir-199 expression levels can be used as novel biomarkers for progression of breast cancer.
