Abstract:

Introduction and ObjectivesAmong amylases, α-Amylase is in maximum demand due to its wide range of applications in the industrial front. Also Amylase has medical applications in the use of Pancreatic Enzyme Replacement Therapy The aim of this study characterization and comparison activity of α-Amylase enzymes that are produced by the three most important α-Amylase producers.Material and methods:All 20 sequences were obtained from NCBI genbank. For computing Physico-chemical properties the Expasy’s ProtParam server and PROTSCALE were used. Secondary structure prediction and Functional characterization analyzed by SOPMA, CYS_REC, Motif Search and SOSUI server. In tertiary structure prediction and structure Validation, we used SWISSMODEL, ERRAT, PROVE, PROCHECK, WHATCHECK and Verify 3D. Active site assessing analyzed with the help of CASTp. Results:Physico-chemical properties, motifs, disulfide bridges, secondary structure, three dimensional structures and catalytic site were predicted and stereo-chemical quality was evaluated by structure validation tools. Results were shown a significant different among three kind of Alpha amylase. Many amino acid changes were detected that have important roles in protein activity and stability. Conclusion:Bioinformatics analysis showed many important properties of Alpha amylase that are applicated in medical industrial application, that it is essential to manipulate the characteristic properties of enzyme.
