Abstract:

Comparative genomics and synteny of MQTL regions is the most accurate ways to identify candidate genes associated with quantitative traits [1]. It was assumed that the wheat chromosome carries yield and it s components related traits gene regions, In addition, the orthologous and homologous regions in barley, rice and brachypodium includes same genomic regions. In the past, 71 MQTLs and 195 flank markers were identified on wheat 21 chromosomes. In this study, 197 EST sequences of molecular markers were extracted by GrainGenes database. Syntenic relationships of wheat-barley, barley-brachypodium and barley-rice genomes were identified by PlantsDB database. Comparative analysis of those MQTLs and also wheat-barley genome alignment were conducted by BLASTn procedures and Gramene database and 57 homologous regions identified. Physical to genetic distances ratio (Kbp/cM) of 57 syntenic MQTLs projected 29 gene rich regions as hot-spots which used for candidate gene identification and 122 candidate genes for 1000 grain weight, grain yield and plant height such as MLOC_68550, MLOC_74389, MLOC_3236, MLOC_38958 and MLOC_61756 were reported. A candidate gene of homologous regions on wheat chromosomes were identified by QuickGo and Gramene databases. Candidate genes can be used to map-based cloning.
