Abstract:
Background: src proto-oncogene codes for a protein-tyrosine kinase that plays key roles in cell growth, division, migration, and survival signaling pathways. The product of the human src gene, c-Src, is found to be over-expressed and highly activated in a wide variety of human cancers including breast cancer. Microsatellites are di-, tri-, or tetra nucleotide tandem repeats in DNA sequences that can exist in promoter, intron or exons of the genes and the number of their repeat can influence the expression of the gene, splicing process or even can change the structure of the final product. Microsatellites can exist in hundreds of locations in the genome and are notable for high diversity in the population. The aim of this study is to find microsatellites in the src gene by bioinformatic studies. Materials and Methods: several databases including NCBI, ensemble, and Human genome browser were used in order to find the sequence of the gene and Dfam database service, were used as tandem repeat finder to find STR markers in the src gene. Result: Among the STRs investigated two STRs, CA repeats in the intron2 and GT repeat in 3’UTR region of the src gene, have been selected as STRs to be investigated in the labrotary . Conclusion: Due to the importance of the src gene in Malignancies, this study has focused on two microsatellites in the intron2 and 3’UTR region of that, to evaluate its association with breast Cancer risk. Finding its relationship with cancer can help us in diagnosing people susceptible to cancer and alsohas therapeutic purposes.
