Abstract:
Introduction & Aim: Breast cancer is the most common invasive cancer in females worldwide. It accounts for 16% of all female cancers and 22.9% of invasive cancers in women. miR-125a-5p promotes cell growth arrest and apoptosis while targeting the proproliferative truncated TrkC isoform. The aim of this study was to compare the expression level of miR-125a-5p in normal and neoplastic samples from breast cancer patients using RT-qPCR.Methods: In the present case-control study, 40 samples from patients with breast cancer tumor and 40 patients from non-tumor were collected under the direct supervision of a pathologist specialist due to clinical presentation and laboratory findings. After extracting RNA from normal and tumor tissues, cDNA synthesis method according to the protocol and RT-qPCR were performed by SYBR®Premix Ex TaqTM II kit. miR-125a-5p expression levels was calculated using ∆∆CT. Data were analyzed using t-test.Results: The results of molecular method indicated that the mean relative expression levels of miR-125a-5p in tumor samples compared to normal was down-regulated. This variation expression of miR-125a-5p in tumor samples was about 3.2 fold less than normal samples.Conclusion: According to previous reports, miR-125a-5p may play an important role in BC progression, and that the down-regulation of miR-125a-5p may serve as independent predictors for breast cancer.
