Investigation biocompatibility and antibacterial properties of chitosan Nano fibers containing silver nanoparticles against Staphylococcus aurous and Pseudomonas aeruginosa

Abstract:

Nosocomial infections are one of the major problems in hospitalized patients and one of the main reasons for the death of these patients, which is one of the main concerns of the World Health Organization due to increased burden on patients and the community. With the increasing consumption of antibiotics across the world and the emergence of resistant strains of common antibiotics and the second line of treatment, numerous studies have shown that the use of nanomaterial‟s(materials with at least one dimension less than 100 nm) are a good alternative for some antibiotics are used to control infections. Today, the development of Nano-dressings is on the agenda of the researchers, so adding nanoparticles alone or with antibiotics can prevent chronic infections in diabetic wounds, burn patients, and even surgical wounds, and speed up the wound healing process. Antibacterial activity of metal nanoparticles has been widely reported. After absorption by bacteria, these materials cause damage to the cell membrane through the release of metal ions, causing the bacteria to die through interventions with proteins and bacterial DNA. In this study, we first studied with anti-bacterial activity of nanoparticles and Nano-scaffolds, chitosan containing silver nanoparticle scaffold was selected and electro spin method was used. Antibacterial effects and MIC (Minimum Inhibitory Concentration) of these nanomaterials were determined by using disk diffusion method and micro dilution techniques against common wound infections with wound infection. The biocompatibility of these nanomaterial‟s was investigated by MTT (MTT test, a color test for the evaluation of metabolic activity of the cells).The results show the effective and optimal activity of nanomaterial‟s against bacteria, especially gram-negative bacteria.
