Abstract:

Background:Polymerase Chain Reaction (PCR) is an important laboratory technique in molecular biology and diagnostics. The design of appropriate short or long primer pairs is only one goal of PCR product prediction. Thesuccess of PCR-based tests mainly refers to primer sequence.In silico PCR searches a sequence database with a pair of PCR primers for fast performance.Method:Determining primer location, orientation, length of each amplicon, simulation of electrophoretic mobility, identification of open reading frames are provided by in silico PCR tools.PCR primer designing is a daily lab activity. Designed primer must be checked if they are applicable to new sequence. Some parameters of PCR experiment can be tracked by in silico PCR. In silico PCR are performed in ugene.unipro.ru and insilico.ehu.es .Results:When the process of in silico PCRis run, the predicted PCR products appear in the products table. Products table includes region of product in the sequence, product length and preferred annealing temperature.Conclusion: In silico PCRis useful computational tools used to calculate theoretical PCR results using a set of primers to amplify DNA sequences from a sequenced genome.It is extremely easy and a cheap way to not get into trouble.
