Abstract:

Introduction: Genome Wide Association Studies (GWAS) showed that risk alleles of chromosome 9P21 region are related to increasing hazard of cardiovascular diseases without any association with their risk factors such as lipid profile and inflammation markers. No study have been performed to evaluate the interaction between these single nucleotide polymorphisms (SNP) and dietary patterns (DP) on cardiovascular disease risk factors. Materials and methods: This cross-sectional study conducted on 265 Tehrani healthy adults (age range: 18-55 years). DPs were extracted by a 147-items semi-quantitative food frequency questionnaire (FFQ) and factor analyzing method. Bioelectrical impedance analysis (BIA) method was used for body analysis. Weight, height, waist circumference, waist to height ratio (WHtR), body mass index (BMI), lipid profile, fasting blood glucose and high sensitivity C-reactive protein (hs_Crp) were measured. Blood sampling was done after 10-12 hours fasting. rs1333048 were genotyped by restriction fragment length polymorphism (PCR-RFLP) method. Results: Prevalence of C allele was 52.85% and A allele was 47.15%. AA genotype compared to CC had greater odds for general obesity (OR=3.11; 95% CI =1.008-9.60, P=0.048) and central obesity (OR=2.63; 95% CI =1.12-6.17, P=0.026). Significant Interactions were observed between legumes based DP and rs1333048 SNP on waist circumference (P-interaction= 0.047), body fat percent (P-interaction = 0.048), hs-Crp (P-interaction =0.042), BMI (P-interaction =0.073), WHtR (P-interaction =0.063) and odds for general obesity (P-interaction =0.051) in this way that last tertile of this DP compared to first tertile, reduced all mentioned items for individuals with CC genotype whereas increased them for who with CA and AA genotypes. Conclusion: Results of this study indicates significant associations between A allele of rs1333048 SNP and general and central obesity and significant interactions between this SNP
