Abstract:

Some male survivors of childhood cancer are in distress from azoospermia. Conventional cancer cell sorting techniques, which have been reviewed elsewhere, are not effective. The aim of this study was to separating the mouse malignant cell line (EL-4) and spermatogonial stem cells in vitro. Spermatogonial cells were co-cultured with mouse malignant cell line (EL-4) cells and divided into 3 culture groups: 1) Control (co-culture in culture medium), 2) Co-culture were sorted by FACS, 3) Co-cultured cells were separated by microfluidic chip. The percent of cells were assayed after treatment using flow cytometry and the identity of cultured cells were confirmed by RT-PCR and immunocytochemistry. The present work demonstrates the use of electrophoretic lab-on-a-chip device in effectively separating cancer cells from spermatogonial cells (42.14 EL4, 0.12 SSC) and (0.32 EL4, 80.38 SSC,) compare with FACS technique.
