EXPRESSION STUDY OF CREB GENE FAMILY IN METHAMPHETAMINE ABUSERS WITH AND WITHOUT PSYCHOSIS

Abstract:

Background and Aim : Methamphetamine abuse alter behavioral characteristic which in some cases includes psychotic symptoms. Mechanisms of psychosis in meth-induced psychosis and its similarities and differences with psychosis in individuals with psychiatric disorders are not completely clarified. Present study aimed to shed lights in role of cAMP response element-binding protein (CREB) a family of transcription factors that are involved in memory and cognition, in methamphetamine abuse and meth-induced psychosis.Methods : Three groups including 20 methamphetamine dependents, 20 meth-induced psychosis and 20 normal non psychiatric subjects were selected. Blood sampling and RNA extraction was operated. Gene expression of CREB family including CREB1, CREB2, CREB3 and CREB4 was evaluated by Real time PCR.Results : Results indicate different expression profiles in Methamphetamine abusers, Meth-induced psychosis in compare with control subjects. Significant over expression of CREB1 and significant down expression of CREB2, CREB3, and CREB4 were detected in both methamphetamine abusers and meth-induced psychosis in compare with normal subjects. mRNA level of CREB1 were over expressed in meth-induced psychosis versus methamphetamine dependent without psychosis. Conclusion : Results suggest that methamphetamine abuse may deregulate genes which are involved in memory formation. In addition CREB family especially CREB1 could be related to meth induced psychosis that is different with psychosis in patients with psychiatric disorders and could considering as biomarkers for meth-psychoses
