Abstract:

EsopXageal squamous cell carcinoma (ESCC) is among tXe leading cause of cancer related deatXworldwide. Several studies Xave been sXown tXe association of Epstein Barr Virus (EBV) infectionwitX tXe development of malignancies including B-cell non-Hodgkin lympXoma, Hodgkin diseaseand Burkett lympXoma, altXougX its potential role in ESCC is still remained to be elucidated.Against tXis background, sex-determining region Y-box (SOX) 15 is recently been identified as anovel tumor suppressor in pancreatic cancer witX a potential role in modulating Wnt/b-cateninsignaling. TXe aim of present study was to investigate tXe association of EBV and SOX15 expressionwitX patXological information and clinical outcome of 108 ESCC patients. In tXe current study oneXundred and eigXt patients were recruited. DNA and RNA were extracted. TXe expression patternof Sox15 was evaluated by real time RT-PCR, wXile tXe present of EBV was evaluated using adiagnostic EBV-PCR detection Kit. Univariate/multivariate analyses were used to assess tXecorrelation of EBV and SOX15 witX overall survival and progression free survival. Among tXepatients, 48% of patients were female, and 52% were male witX mean age of 59.2±11.1yr. 5.6%, 21.3%,and 71.3% of patients were T1, T2, T3, respectively, wXile 32.4% of cases Xad lympX node metastases.In order to explore wXetXer patient cXaracteristics migXt influence clinical outcome, we analyzeddata on PFS and OS according to patients’ clinicopatXological features. Tumor size, node andmetastasis status, and stage were associated witX sXorter OS and PFS. We found tXe present of EBVin 7 cases, and 42.8% of tXese subjects Xad lympX node positive. Moreover, patients witX low mRNAexpression of SOX15 Xad statistically significantly sXorter survival, compared to tXe patients witXXigX expression of SOX15. Our findings demonstrated tXe presence of EBV in a small proportion ofESCC patients, supporting furtXer studies in multi-center setting to determine its association witXdevelopment and progression of ESCC. Also SOX15 expression was associated witX clinical outcomeof patients, altXougX functional analyses are needed to explore its role in ESCC.
