Abstract:
The HOX genes are a highly conserved family of homeodomain-containing transcription factors. HOX genes are associated with a number of malignancies including acute myeloid leukemia. This study wasaimed to evaluate the effect of HOXB1 gene expression in myeloid leukemia samples using Real-Time PCR. A large Iranian kindred with overall 5 affected individuals along with their unaffected siblings andparents were recruited. The candidate gene HOXB1 was screened and analyzed. After RNA extraction from blood, cDNA was synthesized according to protocol and Real-Time PCR carried out using SYBR®Premix Ex TaqTM II kit. HOXB1 expression level was analyzed by ΔΔCT method. The results of this study demonstrate that expression level of HOXB1 gene in homo and hetero had a similarexpression. However, no differences expression level of HOXB1 gene were found. Due to previous studies the HOXB1 mutation involved in acute myeloid leukemia. The number of patients that had 7bpmutation did not show any expression change in HOXB1 gene, probably because the sample mean age was 14 and there is still capable of engaging with the disease. Investigation of HOXB1 gene expression in a larger family might have different results
