Abstract:
Background and aim: Single nucleotide polymorphisms rs8099917 located in 8kb upstream of IL-28B gene. Also it have been confirmed that genetic polymorphisms in the IL-28B gene region are strongly associated with the response to treatment and open the possibility of a personalized medicine for the treatment of the progressive disease. In this study, we investigate set up amplification of rs8099917 fragments .with specific designed primers by RFLP-PCR method Methods: PCR-RFLP technique was used to genotyping of controls. Sequences of SNP rs8099917 were found in Pubmed website and endonuclease restriction enzymes was prepared by NEBcutter tools web- .site. Primers were designed by Gene Runner software Results: In this study, we set up amplify sequences of SNPs fragments by PCR-RFLP for n=79 cases. Fragments of PCR and RFLP were sharp and could be detected genotype of samples (GG/GT/TT) .Conclusion: The PCR-RFLP is simple and useful method for investigation of genotyping of SNPs of IL-28 gene for study samples and can help to treatment. Therefore this method can be enhanced the prediction treatment of some virological patients which can help to clearance
