Abstract:

Introduction: Breast cancer is the second leading cause of cancer deaths among women, and surgical therapies, hormone therapy and chemotherapy are the most commonly used therapies. Hydroxyurea is a chemotherapy drug with side effects such as skin and blood disorders in the treatment of breast cancer. Nowadays, drug nanocariers can increase the effectiveness of anticancer drugs and reduce their adverse effects. Magnetic Chitosan is one of the biocompatible polymer nanocarriers that can be used to target drugs to the tumor tissue in a magnetic field. The purpose of this study is to investigate the cytotoxic effect of hydroxyurea loaded on the magnetic chitosan nanocarrier on breast cancer and to reduce its side effects when directed by the magnetic field.Method: The anticancer effect of magnetic chitosan-hydroxyurea on the breast cancer cell line (MCF-7) by MTT was investigated at 48h and 72h incubation times. In this test, MTT dye is reduced to purple Formazan crystals by living cells. The amount of Formazan which representing percentage of living cells, was calculated by spectrophotometric colorimetric method at 570 nm wavelength.Results: By increasing the incubation time from 48 to 72 hours and increasing the drug concentration from 125 to 2000 µg/ml, the cytotoxic effect of the drug on the MCF-7 cell line increased. So that in 2000 µg/ml nanodrug dose and after 72 hours duration of exposure, the highest mortality rate was observed and 36.6% viability was obtained.
