Abstract:

Multiple sclerosis is an inflammatory demyelinating disease of the central nervous system (CNS) which it hasvariety of symptoms. It can cause muscular weakness, vision problems include double vision, blindness in oneeye and lack of coordination. MS is the most common immune mediated disorder affecting the CNS which MSorganization have estimated that 2.3 million people are living with MS worldwide.However MS is not considered a hereditary disease, a number of genetic variation have been shown to increasethe risk. Some evidences with GWAS (Genome Wide Association Study) confirmed this association. One theassociation is between lncRNA’s (Long Non Coding RNA) mutations and increase the risk of MS.Lncs have gained widespread attention in recent years as potentially new biological regulation. Bioinformaticsand statistical data have showed correlation between some SNPs (Single Nucleotide Polymorphism) and increasethe risk of MS.In this article we have checked the most important associated genes with MS and among them we candidatedIL12A gene (p = 3.08 × 10-9 ). This gene encode interleukin 12 alpha polypeptide that an act as a growth factorfor activated T and NK cells, enhance the lytic activity of NK/ lymphokine-activated killer cells, stimulate theproduction of INF-gamma and showed to play role in pathogenesis of MS. IL12A has almost 9 known lncRNAs,between all of them NONHSAG036523.3 (gene ID) showed association with MS. With bioinformatics analysiswe found the most associated SNPs which include rs2366408, rs4680534 and rs2243123.Together, our studies indicate that the SNPs in NONHSAG036523.3 may play a role in MS pathogenesis andthese are useful for personalized medicine.
