9-tBAP From Spiroaminopyrimidones Family Decreases cell Proliferation and Down-Regulation of Survivin Concomitant withInduction of Apoptosis in NB4 Leukemia Cells

Abstract:
Background and Aim: It has been recently reported the activity ofaminopyrimidone family to induce apoptosis in human cancer cells. Inthis study, we reported an active compound from spiroaminopyrimidonefamily with apoptotic activity against NB4 acute promyelocytic leukemiacells.Methods: The cells were seeded in 96-well plates at 1×105 cells/well andtreated with 10-150 μM of the 2, 4-Diamino-1, 3-diazaspiro [5.5]-9-tertbutyl-2, 4-diene- 5- carbonitrile (9-tBAP). This compound was foundto be highly active cell growth inhibitor with IC50 of 30±3.5 μM asdetermined by MTT assay. Evaluation of survivin expression in NB4 cellstreated with 9-tBAP was performed by real-time PCR.Results: 9-tBAP decreases cell proliferation of the NB4 cells in a doseandtime-dependent manner. The IC50 value following 72 h exposurewas found to be 30 μM for the cells. Furthermore, real-time PCR analysisrevealed that the treatment with the compound down-regulated theexpression of survivin in a time-dependent manner.Conclusion: These data further suggest that 9-tBAP may provide a noveltherapeutic approach for the treatment of leukemia.
