Effects of Trachyspermum ammi essential oil and low level laser irradiation on human breast cancer cells (MDA-MB-231) survival and cell cycle

Abstract:

Purpose: Breast cancer, with an incidence rate of 1.2 million per year, is the most common malignancy (23% of all cancers) in women worldwide. One of the traditional methods used to control the progression of breast cancer is chemotherapy which is often associated with severe side effects. Consequently, in recent years, the role of natural dietary compounds, especially flavonoids, in cancer control and cancer treatment has received much attention from investigators. In the present study, the effect of Trachyspermum ammi essential oil (TEO) alone and in presence of low level laser on MDA-MB-231 breast cancer cells was investigated.Methods: The MDA-MB-231 breast cancer cells were exposed to low level laser irradiation at 660 nm for 90 seconds and then treated with different concentrations of TEO. In another study, cells were treated with different concentrations of TEO initially and then were exposed to low power laser irradiation at 660 nm for 90 seconds. Effect of the low power laser and TEO on cell survival and cell cycle was evaluated by using MTT assay and flow cytometry, respectively. Results: The results showed that early treatment with TEO and then low level laser reduce the survival of breast cancer cells more than the early treatment with low level laser irradiation then essential oil. Also, TEO treatment at IC50 concentration in combination with low level laser irradiation induced G0/G1 cell-cycle arrest in MDA-MB-231 cells as compared to the dark group.Conclusion: This study showed that the Trachyspermum ammi essential oil in combination with low level laser results more reduction in the survival and lead to cell cycle arrest in MDA-MB-231 cells.
