Abstract:

There are various scaffold have been fabricated based on materials ranging from naturally to artificial synthetically. We hypothesized that a biological scaffold derived from deposited extracellular matrix of keratinocytes on collagen can improve cutaneous wound healing. With regard to these considerations, the current study aimed to fabricate a biological scaffold by culture of keratinocytes in 3D collagen scaffold. The keratinocytes were cultured in 3D collagen scaffolds in keratinocyte medium, supplemented with 10% (v/v) FBS, 10 μg/mL streptomycin and 100 mg/mL penicillin for a week. The cell were made extracellular matrix and added to collagen during a week. Afterward decellularization and lyophilization has been done on 3D matrixes. Glycosaminoglycans (GAGs) was identified in extracellular matrix of cells by safarin o and alcian blue staining. All the scaffolds were evaluated by scanning electron microscope and FTIR.Fourier transforms infrared spectrometry (FTIR) and scanning electron microscopy (SEM) analysis identified that this scaffold are highly porous with interconnection. This can provide an ideal scaffold which is suitable for cell growth and differentiation. This 3Dporous scaffolds showed a different physical and biological property that is useful in tissue engineering applications in wound healing.
