Introduction & Aim: Esophageal Cancer (EC) has been noticed to lethal malignancy and lower survival rate. Hypoxia-Inducible Factor 1 Alpha (HIF-1A) is considered in the center of solid tumor tissue because of hypoxia condition. Dysregulation of HIF-1A and its downstream pathway have a critical role in tumorigenesis process in different cancers. The expression of HIF-1Ain EC patients have been evaluated with respect to association with EC tumor stage. Methods: This investigation conducted on ESCC patients (n=40). The expression level of HIF-1A evaluated in tumor tissues and their adjacent normal tissues in EC patients. Primarily, total RNA extracted from each sample separately. Then RNA was use for, cDNA synthesis and expression level of HIF-1A evaluated by quantitative Real Time PCR. GAPDH was used to normalized the expression level of HIF-1A. The association between these genes expression with clinical parameters in EC patient were analyzed. Results: HIF-1A was markedly upregulated in EC tumor tissues compared with non-tumor tissues. A high level of HIF-1A expression was associated with advanced stage (p<0.001). Conclusion: In this present study HIF-1A showed higher expression in human esophageal cancer. HIF-1A is an essential factor for tumor progression and metastasis in various cancer. HIF-1A is a useful marker for predicting patient outcome and could be applied for planning new therapeutic strategies for EC.
