Background: The minor A-allele of rs12970134 in intronic region of melanocortin 4 receptor gene (MC4R) is one of the most famous allele in related with body mass index in GWA studies. In presentstudy, we reported replicating finding in a large Iranian population-based cohort study, Tehran lipidand glucose study (TLGS). Methods: Among TLGS population 1777 cases with metabolic syndrome (MetS) according to four definitions ATP III, AHA, IDF and JIs, also 2170 Diabetic case, were selected and compared to … control. This polymorphism was genotyped by using the Centaurus (Nanogen) platform in DeCODEgenetics. By using plink software, allelic association with metabolic syndrome and diabetic patientsalso related components (e.g. lipid profile, waist, BMI and sugar profile) were analyzed, after age and sex adjustment.Results: the outputs clearly showed A-allele of rs12970134 had associated with three definition of fourdefinition of MetS prevalence (p< 0.0065) in ATP III with highest level as well as diabetes (p <0.0049) and most related components. Conclusion: Our findings showed A-allele of rs12970134 among Iranian population surplus waistcircumference and related phenotypes confirmed previous result in other ethnicity have association with MetS and diabetes and related traits.
