Abstract:

The optimization of the kinetic parameters is discussed and derived by simulating the antibody-antigen reaction in a microfluidic channel. A model is designed and used in COMSOL multiphysics with a 2D geometry to optimize the kinetics. The convectiondiffusionequations combined with diffusion-reaction equations develop a model to investigate the antibody-antigen complexes' concentration behavior. The monoclonal antibody (MAb) was immobilized on the microchannel wall surface, and the prostate specific antigen (PSA) was directed through the channel by the analyte solution. Theeffect of velocity and coated antibody was determined via a wide range of plots. Then the optimized velocity and antibody concentrations were derived after analyzing the data. Generally, the complex concentration rises and reaches equilibrium by increasing the velocity and decreasing the immobilized antibody concentration. The 2D simulation exhibited an excellent prediction for optimizing and obtaining accurate results.
