Abstract:
این تحقیق به منظور اعتبارسنجی تعداد 10 نشانگر ریزماهواره پیوسته با ژن‌ها و جایگاه‌های مختلف تحمل شوری از جمله ژن Saltol که به فاصله صفر تا هشت سانتی‌مورگان از جایگاه‌های ژنی قرار داشته و ارزیابی تنوع آللی این ژن‌ها در 59 ژنوتیپ
برنج طی سال‌های90-1388 در موسسه تحقیقات برنج کشور انجام شد. در این آزمایش ارزیابی برای تعدادی از صفات مورفولوژیک مهم مرتبط با تحمل تنش شوری شامل نمره تحمل شوری، ارتفاع گیاهچه، وزن خشک ریشه، وزن خشک اندام هوایی و محتوای آب نسبی برگ انجام شد. ژنوتیپ‌های برنج در مرحله گیاهچه‌ای در دو تیمار 9/0 دسی‌زیمنس بر متر (بدون تنش)
و 12 دسی‌زیمنس بر متر (تنش شوری) مورد ارزیابی قرار گرفتند. غربال مولکولی با استفاده از 10 نشانگر مورد استفاده در ژنوتیپ‌های برنج مورد مطالعه، 60 آلل را در ژنوتیپ‌های برنج ایجاد نمود. نتایج حاصل از تجزیه رگرسیون لجستیک نشان داد که از 10 جایگاه ریزماهواره پیوسته، چهار نشانگر توجیه‌کننده تغییرات در واکنش به شوری به‌ترتیب RM315 برای ارتفاع گیاهچه، RM223 برای وزن خشک ریشه و ارتفاع گیاهچه، RM3627 برای وزن خشک ریشه، ارتفاع گیاهچه و محتوای آب نسبی برگ و RM8094 برای برای وزن خشک ریشه و محتوای آب نسبی برگ بودند. بر اساس تجزیه خوشه‌ای براساس داده‌های مولکولی با استفاده از ضریب Yule و روش UPGMA، ژنوتیپ‌های برنج به پنج گروه تقسیم شدند. با توجه به اینکه سه نشانگر پیوسته شناسایی شده (RM8094 ، RM3627 و RM315) روی کروموزوم یک واقع شده و با ژن Saltol نیز پیوسته هستند، می‌توان از آنها در شناسایی ژنوتیپ‌های متحمل و حساس برنج استفاده کرده و ژنوتیپ‌های واجد این جایگاه‌ها را در برنامه‌های اصلاح برای تحمل به شوری مورد استفاده قرار داد.
