Cytotoxicity analysis of diazinon on HEK-293 cell using MTT assay

Diazinon is an organophosphate pesticide, and nowadays this pesticide is mostly used in agriculture. In this investigation, we studied the toxic effects of diazinon in HEK-293 cells using MTT assay. A range of diazinon concentrations (5, 50 and 500 μM) were treated to analyze cytotoxicity. 24 hours after exposure, MTT assay was used for cytotoxicity evaluation. Diazinon induced some morphological changes such as vacuolization, cell rounding and cell blebbing. In addition, diazinon reduced the viability of HEK-293 (82±3%), (51±4%) and (15±2%) at 5 μM, 50 μM and 500 μM (P<0.05), respectively. To sum up, diazinon is cytotoxic for HEK-293 cells in dose-dependent pattern.
