Abstract:
Background and Objective: Ecto-nucleotide pyrophosphatase/phosphodiesterase ۱ (ENPP۱) is a class II membrane glycoprotein that binds to insulin α receptor and can interfere in insulin signaling pathway. Transcription factor-۷-like ۲ (TCF۷L۲) is a transcription factor which plays a critical role in pancreatic β cell activity. ENPP۱ and TCF۷L۲ gene polymorphisms may have functional role in susceptibility to type۲ diabetes (T۲D). The aim of this study was to investigate the association of reported K۱۲۱Q and C/T rs۷۹۰۳۱۴۶ variants of ENPP۱ and TCF۷L۲ genes with the risk of T۲D in our population
Material and Methods: ۲۴۰ T۲D and ۲۴۰ healthy subjects were recruited. Genotyping was carried out by PCR-RFLP method. T test was used for association study.
Results: ENPP۱ ۱۲۱Q (CC) genotype was significantly higher in T۲D comparing to controls (OR;۱.۶۱, ۹۵% CI;۱.۰۲-۲.۵۵, P=۰.۰۲) and a significant association between the frequency of C allele and T۲D was observed (OR;۱.۳۳۹, ۹۵%CI;۱.۰۴-۱.۷۲, P=۰.۰۱۲). TT genotype of TCF۷L۲ C>T rs۷۹۰۳۱۴۶ was significantly higher in T۲D patients (OR;۰.۶۷, ۹۵% CI;۰.۴۹-۰.۹۸, P=۰.۰۲), but the T allele could not significantly affect the risk for T۲D in our population.
Conclusion: The high frequency of Q allele of the ENPP۱ K۱۲۱Q and TT genotype of the TCF۷L۲ might be considered as a predisposing factor for T۲D.
