Microarray data analysis and Association between IL-۲۷ rs۱۵۳۱۰۹ Polymorphism and the Risk of Bladder cancer in a sample of Iranian population

Abstract:

Background and Aim: Bladder cancer is considered the fifth most common cancer worldwide and it is associated with significant morbidity and mortality. increasing age, male gender, cigarette smoking, chronic inflammation due to infection, exposure to chemical carcinogens such as aromatic amines and genetic factors have been identified as risk factors. Interleukin (IL)-۲۷, a novel member of the IL-۱۲ family, inhibits growth of cancer cell. The present study aimed to assess the impact of IL-۲۷ rs۱۵۳۱۰۹ on bladder cancer in a sample of Iranian population.Methods: The Agilent single channel expression profile by array with accession numbers GSE۹۴۴۲۴ was downloaded from GEO Datasets of NCBI database and it is analyzed with R statistical language using Limma package. By using GeneFiltration, genes are filtered and quality control performed before and after normalization then differential expression genes determined. Then the most significant gene is chosen for the case-control study on ۱۴۰ bladder cancer and ۱۴۲ controls. DNA was extracted from whole blood by salting out method and evaluation of genetic polymorphism was performed using PCR – RFLP method. Chi-square, independent sample t-test and logistic regression analysis were used to determine Statistical analyses.Results: The microarray analysis has shown interleukin ۲۷ gene expression significantly reduced in tumor cells compared to control samples (logFC= -۶.۲۴, p.value=۰.۰۴). our findings on single nucleotide variation show that IL-۲۷ rs۱۵۳۱۰۹ TC genotype and C allele of IL-۲۷ decreased the risk of bladder cancer. (OR= ۰.۳۴, ۹۵% CI= ۰.۲۰-۰.۵۷, P<۰.۰۰۰۱ and OR= ۰.۶۳, ۹۵% CI= ۰.۴۴-۰.۹۰, P=۰.۰۱۱, respectively).Conclusion: IL-۲۷ serves as an anti-tumor factor by inhibiting angiogenesis with CLCX۹ and CLCX۱۰ as mediators. Our study confirmed IL-۲۷ downregulated in cancer samples compared to healthy individuals also, shows that IL-۲۷ rs۱۵۳۱۰۹ variant may have a protective role in control samples compared to those with bladder cancer in the sample population.
