Abstract:

Background and Aim: Stemness biomarkers including Oct-۴, Nanog and Sox-۲ are considered to be responsible for tumor relapse and metastasis. Here, we aimed to study the effects of Gemini Curcumin on the expression of cancer stem cell markers in HCT۱۱۶ colon cancer cell line.Methods: HCT۱۱۶ cells were treated with Gemini Curcumin and free Curcumin in a time and dose-dependent manner. Cellular toxicity was studied by using MTT assay and cell cycle analysis. Real time PCR and western blotting were employed to evaluate gene expression.Results: our data showed that Gemini-Cur significantly modulates HCT۱۱۶ cells prolifration. Furthermore, qPCR and western blotting showed that Oct-۴, Nanog and Sox-۲ are significantly down-regulated in Gemini-Cur treated cells compared to control.Conclusion: Collectively, the data demonstrates the therapeutic potential of Gemini Curcumin against cancer stem cell markers that play role in cancer relapse and metastasis.
