Association study between polymorphisms in MIA۳, SMAD۳, SELE, CETP, LIPG and tRNAThr genes, and Coronary Artery Disease (CAD)

Abstract:

Background and Aim: One of the main reasons of mortality in the world is cardiovascular diseases, and coronary heart disease (CAD) is the most common heart disease that occurs as a result of coronary artery stenosis and atheroma plaque formation. According to the definition of World Health Organization, CAD refers to cases with ≥۵۰% luminal stenosis of any coronary vessel. CAD is a multifactorial disease which genetic and environmental factors are involved in it. Several studies has shown association between some polymorphism in different genes with CAD. Finding this association can be used in order to early diagnosis and prevention of CAD.Methods: ۱۰۰ CAD patients with ≥۵۰% luminal stenosis of any coronary vessel as case group and۱۰۰ healthy individuals as control group were selected. After DNA extraction by a standard extraction kit, the polymorphisms were evaluated by ARMS-PCR and PCR-RFP methods. Sanger sequencing method was used to confirm the findings. In this study association between rs۱۷۲۲۸۲۱۲ in SMAD۳ gene, rs۱۷۴۶۵۶۳۷ in MIA۳ gene,rs۳۹۱۷۴۰۶, rs۱۸۰۵۱۹۳ and rs۵۳۵۵ in SELE gene, rs۵۸۸۲ and rs۷۰۸۲۷۲ in CETP gene, rs۲۰۰۰۸۱۳ in LIPG gene, rs۱۹۳۳۰۳۰۰۲ in tRNAThr and the risk of CAD were evaluated.SPSS was used for statistical analysis. Frequency of different genotypes of the polymorphism was analyzed using chi-squared (χ۲) test.Results: The results of this study show that there is no significant association between rs۱۷۲۲۸۲۱۲, rs۱۷۴۶۵۶۳۷, rs۱۹۳۳۰۳۰۰۲, rs۱۸۰۵۱۹۳, rs۷۰۸۲۷۲ and rs۲۰۰۰۸۱۳ and risk of CAD in the studied patients. But there is significant association between rs۵۳۵۵ (p=۰.۰۳), rs۳۹۱۷۴۰۶ (p=۰.۰۲), and rs۵۸۸۲ (p=۰.۰۰۷), and risk of CAD.Conclusion: Our results, according to previous studies confirmed although there was no direct association of CETP activity with CETP polymorphisms, our findings revealed a significant interaction between CETP SNPs and CETP activity for affecting HDL-C levels. The SELE gene is a known cell adhesion molecule with a significant role in inflammation. Mounting evidence has suggested inflammation as an important component in the pathological process of coronary atherosclerosis. Studies about possible linkage between SELE gene polymorphisms and the development of CAD are conflicting. We have found a significant association between polymorphisms of SELE gene and risk of CAD.
