Abstract:

Background and Aim: Brain cancer or brain tumor is an abnormal growth of the brain or is a type of hard intracranial neoplasm and intracerebral or central canal of the spinal cord, which may be benign or malignant. Glioblastoma multiforme is the most common and malignant primary brain tumor and usually occurs in the ۶th to ۷th decades thiosemicarbazones complexes are compounds with a long history and a wide range of biological properties including anticancer, antibacterial and anti-malarial effects. Currently, the main known effects associated with their anticancer activity include: Reactive oxygen production (ROS), topoisomerase II inhibition, and more recently inhibition of multiple drug resistance protein (MDR۱). The purpose of this study was to Evaluation of alternations LncRNA CACS۲ expression in the T۹۸G Glioblastoma cell line under treatment with thiosemicarbazones complexes(Cu)Methods: In this study, thiosemicarbazones complexes(Cu) were prepared with ۱۷.۵ and ۲۰ µM concentrations. The adenocarcinoma T۹۸G cell line was treated by Cu in these groups after cell passage. Then RNA extraction and cDNA synthesis were performed and the expression of LncRNACACS۲ in the PI۳ Kinase / AkT signaling pathway and GAPDH gene as the housekeeping gene were evaluated by Real Time PCR. Statistical analysis was performed.Results: Our results confirmed that, at both concentrations of ۱۷.۵ and ۲۰ µM a considerable gene expression increased observed after ۲۴ hours treatment.At ۱۷.۵ and ۲۰µM concentrations, expression was detected ۰.۵۶۴ and ۰.۲۳۴ fold respectively.Conclusion: The results of trials related to thiosemicarbazones complexes Cu and comparison with control drug showed that Cu can be effective in increasing LncRNA CACS۲ expression as a tumor suppressor.
