How Runx-۲ gene is expressed in Sinusoidal Electromagnetic Field pretreated bone marrow-derived mesenchymal stem cells

Abstract:

Background and Aim: Bone marrow-derived mesenchymal stem cells (BMSCs) are multipotent cells with self-renewal and potential to differentiate into osteoblasts, chondrocytes and adipocytes are the promising tool in tissue engineering and regenerative medicine. The stimulatory and inhibitory effects of electromagnetic field (EMF) depend on frequency, field intensity and duration of exposure. The effect of EMF on osteogenesis and expression of Runx-۲ has been reported. This gene is an important transcription factor that regulates osteoblast maturation. In this study, the expression of Runx-۲ was evaluated in sinusoidal EMF (SEMF)-induced BMSCs.Methods: Identification of third passage of isolated mesenchymal from Wistar rats were confirmed by flow cytometry and then divided into three groups, including negative control cultured in a-MEM and ۱۰% FBS, positive control cultured in bone differentiation medium, treated cells exposed to SEMF with intensity of ۱ mT and frequency of ۵۰ HZ, half an hour per day for ۱۴ days. The expression of Runx-۲ gene was evaluated by Real-time PCR.Results: BMSCs had positive response to CD۷۳, CD۹۰, CD۱۰۵ markers and negative response to CD۴۵ marker and differentiated into adipocytes and osteocytes. SEMF-exposed cells showed a significant decrease of gene expression compared to negative and positive control groups.Conclusion: EMF can be applied to the injured bone at the appropriate frequency and at ۵۰ HZ frequency and ۱ mT intensity was not effective in differentiation of BMSCs to osteoblasts.
