Background: Breast cancer is the most common cause of cancer-related deathin women worldwide. Novel genetic markers for breast cancer susceptibility havebeen identified in population-based studies. The aim of this study was to examinethe association of two single-nucleotide polymorphisms (SNPs) of FGFR۲(rs۱۲۱۹۶۴۸) and TOX۳ (rs۸۰۵۱۵۴۲) with the risk of breast cancer in Iranian women.Methods: Breast cancer patients (n = ۱۲۶) and healthy controls (n = ۱۶۰) weregenotyped for SNPs in FGFR۲ (rs۱۲۱۹۶۴۸) and TOX۳ (rs۸۰۵۱۵۴۲) using the tetraprimeramplification refractory mutation system-polymerase chain reaction(ARMS-PCR). Also, immunohistochemical tests for human epidermal growthfactor receptor-۲, estrogen receptor, and progesterone receptor were carried out onbreast tumor tissues.Results: TOX۳ (rs۸۰۵۱۵۴۲) CC (OR = ۱.۲۴; ۹۵% CI, ۰.۷۲-۰.۲۱۴; P < ۰.۰۰۱)and FGFR۲ (rs۱۲۱۹۶۴۸) GG (OR = ۶۲.۰; ۹۵% CI, ۲۳.۶۳-۱۶۲.۶۶; χ۲ =۱۳۲.۷۷۵ ;P < ۰.۰۰۱) polymorphism was significantly associated with breast cancer. Theassociation was also significant between breast cancer risk and TOX۳ (rs۸۰۵۱۵۴۲)TC and FGFR۲ (rs۱۲۱۹۶۴۸) AG variants.Conclusion: Our findings suggested that genetic variants of FGFR۲ (rs۱۲۱۹۶۴۸AG) and TOX۳ (rs۸۰۵۱۵۴۲ TC) can be potential candidate biomarkers for breastcancer risk.
