Common Variations in Perilipin rs۱۰۵۲۷۰۰ and FTO rs۳۷۵۱۸۱۲ Genes, Central Obesity, and Risk Type-۲ Diabetes

Abstract:

Background: Perilipins are proteins at the external level of the lipid glob in adipocytes steroid-generating cells and play a central role in lipid storage and breakdown. FTO gene is associated with Type-۲ diabetes (T۲D) and increased fat mass. Association of Perilipin and FTO genes polymorphism with T۲D was investigated. Methods: Clinical traits in a random sample of ۱۸۳ Iranian men and women with T۲D and ۱۷۴ controls without any metabolic disease took part in the study. The sample was genotyped using Tetra-ARMS method. Results: In Perilipin (rs۱۰۵۲۷۰۰) polymorphism tested, a significant association was found between this polymorphism and T۲D (P=۰.۰۳). In FTO (rs۳۷۵۱۸۱۲) polymorphism, a significant association was seen between this gene and obesity (P=۰.۰۰۳). But, no significant relation was found between this genotype and T۲D. Conclusion: The rs۳۷۵۱۸۱۲ polymorphism of FTO gene is the major genetic determinant of obesity, but not type-۲ diabetes. The rs۱۰۵۲۷۰۰ polymorphism of Perilipin gene is related to T۲D but not obesity.
