In silico study to survey the active ingredients' effects of medicinal herbs on inhibition of Interleukin ۶ receptor to suppress inflammation caused by the cytokine storm in COVID-۱۹

Abstract:

SARS-CoV-۲ has caused a huge problem worldwide and many casualties. Severe inflammation and destruction of the lungs is a major cause of death in patients with COVID-۱۹, and IL۶ plays a key role in the progression of the disease by triggering cytokine storms. Therefore, blocking the IL۶ receptor can be a therapeutic target. The names of plants affecting infectious diseases were achieved from various articles,then, the PubChem database was used to obtain the ۳D structure of their active ingredients. The IL۶Rstructure was also obtained using PDB and NCBI databases and the Swiss Model server. Molecular dockingwas performed on ۱۱۴۵ active ingredients with IL۶R using Auto dock Vina software version ۴.۲.۶ andChimera version ۱.۱۴. Among the active substances, we identified five substances that had the highestaffinity for the receptor. Thus, the Wey۲drug, LAZAR, and AVCpred databases were used to examine thedifferent properties of these substances. Five active ingredients that had the highest affinity for interactionwith IL۶R were Berbamine, Hypericin, Protohypericin, Pseudohypericin, Naphthalene, and ۲- (۱-adamantyl) -۷- (۲-adamantyl) from plants named Mahonia aquifolium, Berberis vulgaris, Hypericumperforatum, Hibiscus sabdariffa, and Lawsonia inermis, respectively. It can be concluded that using theseeffective substances can prevent cytokine storms and chronic inflammation caused by IL۶ in COVID-۱۹.It is hoped that in the future, with further comprehensive bioinformatics research and laboratory studies ofthese five substances, an effective step will be taken in the treatment process of patients with Covid-۱۹.
