Multi-component reactions (MCRs) enable the facile, automated and high throughput generation of small organic molecules. MCRs have been used to create diversity oriented and biased combinatorial libraries, to accomplish the synthesis of highly complex natural products as well as for the large-scale production of drug candidates. This provides medicinal chemists with a powerful tool to create novel chemical diversity, matching the space of biological targets with relevant chemistry. A facile and rapid multicomponent synthesis of ortho-aminocarbonitrile tetrahydronaphthalenes has been developed from benzaldehyde, malononitrile, and cyclohexanone. Different method from the same reactants, solvent, temperature, and catalyst lead to one product with excellent yields. The reaction were followed with the Michael addition and cyclization. The development of a viable catalytic process for industrial scales is a complex task that requires the collaboration of multiple disciplines. In this study, ionic liquid-tethered colloidal silica nanoparticles that increases the yields of products and decreases the time of reactions. For characterization of catalyst some method such as scanning electron microscopy (SEM), energy dispersive X-ray analysis (EDX), transmission electron microscopy (TEM) and X-ray diffraction (XRD) were used.
