Corilagin potential in inhibiting oxidative and inflammatory stress in LPS-induced murine macrophage cell lines (RAW ۲۶۴.۷)

Abstract:

Objective(s): Inflammation is thought to be the common pathophysiological basis for several disorders. Corilagin is one of the major active compounds which showed broad-spectrum biological and therapeutic activities, such as antitumor, hepatoprotective, anti-oxidant, and anti-inflammatory. This study aimed to evaluate the anti-oxidant and anti-inflammatory activities of corilagin in LPS-induced RAW۲۶۴.۷ cells.Materials and Methods: Anti-oxidant activities were examined by free radical scavenging of H۲O۲, NO, and *OH. The safe concentrations of corilagin on RAW۲۶۴.۷ were determined by MTS [۳-(۴,۵-dimethylthiazol-۲-yl)-۵-(۳-carboxymethoxyphenyl)-۲-(۴-sulfophenyl)-۲H-tetrazolium] assay on RAW۲۶۴.۷ cell lines. The inflammation cells model was induced with LPS. The anti-inflammatory activities measured IL-۶, TNF-α, NO, IL-۱β, PGE-۲, iNOS, and COX-۲ levels using ELISA assay.Results: The results showed that corilagin had a significant inhibition activity dose-dependently in scavenging activities toward H۲O۲, *OH, and NO with IC۵۰ values ۷۶.۸۵ µg/ml, ۲۶.۶۸ µg/ml, and ۶۶.۶۴ µg/ml, respectively. The anti-inflammatory activity of corilagin also showed a significant decrease toward IL-۶, TNF-α, NO, IL-۱β, PGE-۲, iNOS, and COX-۲ levels at the highest concentration (۷۵ µM) compared with others concentration (۵۰ and ۲۵ µM) with the highest inhibition activities being ۴۸.۰۹%, ۴۲.۳۷%, ۶۵.۶۹%, ۲۶.۴۷%, ۴۶.۸۸%, ۵۶.۲۲%, ۵۹.۹۹%, respectively (P<۰.۰۵).Conclusion: Corilagin has potential as anti-oxidant and anti-inflammatory in LPS-induced RAW ۲۶۴.۷ cell lines by its ability to scavenge free radical NO, *OH, and H۲O۲ and also suppress the production of proinflammatory mediators including COX-۲, IL-۶, IL-۱β, and TNF-α in RAW ۲۶۴.۷ murine macrophage cell lines.
