Introduction: Ovarian cancer is the fifth most lethal cancer in women. According to the low efficiency of common therapies and also high rate of cancer recurrence, developing a new effective therapy is greatly in demand. Natural Killer Cells and Natural Killer Cell derived Extracellular Vesicles (NK-EVs) have therapeutic potential in cancer therapy. Therefore, in this study we have evaluated cytotoxic effect of natural killer cell derived extracellular vesicles on SKOV۳ cells in ovarian cancer. Methods: First, NK cells were activated by adding IL۱۵ (۱۰ ng/ml). Then, culture supernatant was harvested after ۲۴ h by centrifugation at ۲۰۰۰ rpm for ۵ min to remove cells. Different fractions of NK-EVs were isolated by differential centrifugation at ۲۰۰۰g, ۱۰۰۰۰g and ۲۰۰۰۰g respectively. SKOV۳ cells were treated with NK-EVs. Cytotoxicity of the NK-EV was evaluated using MTS assay. The study groups were ۲۰۰۰g NK-EV treatment group, ۱۰۰۰۰g NK-EV (large EV/lEV) treatment group, ۲۰۰۰۰g NK-EV (medium EV/mEV) treatment group, NK cell derived conditioned medium and control group. Results: According to MTS results, mEVs have the highest cytotoxic effect on SKOV۳ cells. Conclusion: NK-EVs have cytotoxic effect on SKOV۳ cells in vitro. NK-EV is considered as an off-the-shelf product and easy to use and store for a long time. Therefore, it could be a great cancer therapy strategy in the future.
