Abstract:

The anxiety-like behavior resulting from acute diazinon (DZN) exposure has been reported. In this study acute neurotoxicity was induced with single dose of DZN ۱۰ mg/Kg for ۲۴ hours in adult male rats. Behavioral experiments on the elevated plus maze (EPM) and open field (OP) were investigated for anxiety measurement following DZN and/or the pegilated liposomes (QPEGL) ۲۰ mg/kg treatment after ۲۴ hours. Animals were decapitated, brains removed, and frozen at -۸۰. The brain serotonin receptor (HTR۱A) expression was determined by qRT-PCR. The total motor activity level was considered as an index for anxiety measurement. In the EPM results, total motor activity in the DZN group decreased than the control not significantly (P > ۰.۰۵) this parameter in the QPEGL+DZN group had no different than the control one (P > ۰.۰۵) but was significant than the DZN one (P < ۰.۰۵). In open field results total motor activity parameters significantly decreased in the DZN group than the control one (p < ۰.۰۵), in the QPEGL+DZN group, had no significant difference than the control group (P > ۰.۰۵) and increased than the DZN group insignificantly (P > ۰.۰۵). The expression of HTR۱A in the DZN group reduced than the control one differently (P < ۰.۰۵), in the DZN+QPEGL group had no different than the control one (P > ۰.۰۵) and upgraded than the DZN group insignificantly (P > ۰.۰۵). In conclusion, we observed that acute low-level DZN exposure can lead to anxiety-like behavior and has a significant effect on reducing serotonin receptor expression that is improved by QPEGL.
