Abstract:

A novel paradigm in tumor biology suggests that Non-Small Cell Lung Cancer (NSCLC) growth is drivenby lung cancer stem cell-like cells (LCSCs). SLC۳۴A۲, a member of the solute carrier gene family, encodesNaPi۲b (the type II Na/Pi co-transporter). NaPi۲b is a multi-transmembrane sodium-dependent phosphatetransporter responsible for transcellular inorganic phosphate absorption, maintenance of phosphatehomeostasis, and tumorigenesis. The biological function of SLC۳۴A۲ and its underlying mechanisms inNSCLC has remained unclear. In this study, the SLC۳۴A۲ gene was the target for bioinformatics findings.For this purpose, single nucleotide polymorphism (SNP) and microRNA (miRNA) associated with SLC۳۴A۲were found in the GeneCards and NCBI databases. LncRNADisease (version ۲.۰) database helped to analyzethe data to find a long non-coding RNA (lncRNA) related to same miRNA. The analysis showed that theoccurance of the SNP:rs۵۹۹۶۳۹۷ can lead the cells to lung cancer and affect the involved miRNA-۶۵۰ andlncRNA-MEG۳ in this process. The rs۵۹۹۶۳۹۷ occures at the seed match of miRNA-۶۵۰ on ۳’UTR ofSLC۳۴A۲ gene. According to findings from the GEPIA۲ database, the SLC۳۴A۲ gene is highly expressedin lung cancer. Through the process of this disease, the disease-associated SNP: rs۵۹۹۶۳۹۷ is located on themRNA and causes the disease. When miRNA-۶۵۰ binds to ۳’UTR of SLC۳۴A۲ mRNA, it increases geneexpression and intensifies the tumorigenesis of LCSCs. When lncRNA-MEG۳, which is a tumor suppressor,binds to miRNA-۶۵۰ and suppresses SLC۳۴A۲, it silences miRNA-۶۵۰ and reduces pathogenicity. based ondifferent aspects of this pathway, it is inferred that the interaction of lnc-MEG۳ with rs۵۹۹۶۳۹۷ could beuseful biomarkers for the diagnosis of lung cancer. In conclution, the miRNA-۶۵۰ prove that SLC۳۴A۲ is aneffective gene in lung cancer outbreak and its tumorigenesis.
