Abstract:

Visual one-step simultaneous detection of low-abundance methylation is crucial challenge inearly cancer diagnosis in simple manner. Through the design of closed split bipolarelectrochemistry system (BE), detection of promoter methylation of tumor suppressor genes inpapillary thyroid cancer, RASSF۱A and SLC۵A۸, was achieved using electrochemiluminescence.For this purpose, electrochemiluminescence of luminol loaded into the Fe۳O۴@UiO-۶۶ and goldnanorods-functionalized graphite-like carbon nitride nanosheet (AuNRs@C۳N۴ NS), separately,on the anodic and cathodic pole bipolar electrode (BPE) in two different chambers of bipolar cellwere recorded on a smartphone camera. To provide same electric potential (ΔEelec) through theBPEs to conduct simultaneous light emission,as well as to achieve higher sensitivity, anodic andcathodic poles BPEs was separately connected to ruthenium nanoparticles electrodeposited onnitrogen-doped graphene-coated Cu foam (fCu/N-GN/RuNPs) to provide hydrogen evolutionreaction (HER) and polycatechol-modified reduced graphene oxide/pencil graphite electrode(PC-rGO/PGE) to provide electrooxidation of hydrazine. Moreover, taking advantages of strongcathodic ECL activity due to the roles of AuNRs, as well as the high density of capture probe onthe UiO-۶۶ and Fe۳O۴ role in improving the signal-to-background ratio (S/B) in complicatedplasma media, a sensitive visual ECL immunosensor was developed to detect two different genesas a model target analyte in patient plasma samples. The ability of discrimination of methylationlevels as low as ۰.۰۱% and above ۹۰% clinical sensitivity in thyroid cancer patient’s plasmaimplies the present strategy is able to diagnose cancer early as well as monitoring responsespatients to therapeutic agents.
