Abstract:

Hydrogel wound dressings with good biocompatibility and swelling capability are increasingly used in skin tissue engineering applications. Platelet concentrates such as platelet-rich fibrin (PRF) have also received a great deal of attention in recent years for use in wound treatments due to release of different types of growth factors. The composition of PRF in a hydrogel matrix for use as a wound dressing is a less studied topic. In this study, sponge hydrogel dressings were prepared from chitosan/polyethylene glycol (CsPEG) with different percentages (۱, ۱.۵ and ۲% w/v) of advanced PRF (A-PRF). Based on results, CsPEG sponge contains ۱.۵% w/v of A-PRF (CsPEG/۱.۵A-PRF) had the highest mechanical properties (tensile strength, elastic modulus and elongation at break) among all sponges and was therefore selected as the optimal sample for other tests. CsPEG/۱.۵A-PRF sponge also showed the most intense antibacterial activity against E. coli and S. aureus compared with other sponges. Enzyme-linked immunosorbent assays revealed that PDGF-AB and VEGF had an incrementally release from CsPEG/۱.۵A-PRF sponge in ۷ days. The results of MTT assay also showed that none of the prepared sponge dressings were cytotoxic and CsPEG/۱.۵A-PRF sample significantly increased L۹۲۹ cells proliferation and viability in ۵ days compared to other samples. Eventually, the results of this study introduce CsPEG/۱.۵A-PRF sponge as a suitable candidate for skin tissue engineering applications.
