CAR NK cell therapy and breast cancer

Breast cancer is the most frequentmalignancy in women. Besides standardtreatment approaches (surgery, chemotherapy,radiotherapy and hormonal blockade), newCAR T cell therapy as an adoptiveimmunotherapy appears a promising strategyto control the advanced stages of cancer byspecifically targeting the tumor. There areseveral clinical trials of CAR-T cellstargeting HER-۲ in progress, such as aphase I/II study of HER-۲- targeted CAR-Tcells in chemotherapy or HER-۲ antibodyinhibitor therapy for refractory HER-۲-advanced breast cancer.CAR-T cells are synthetic molecules inwhich the effector function of Tlymphocytes combines with the ability ofantibodies to identify specific antigens.CAR molecules can be divided into ۳components: an extracellular domain,which is involved in antigen identification,transmembrane domain and intracellulardomain, which is plays a fundamental role in signal transduction aimed at activating Tcells. CAR T cell therapy has beensuccessfully applied to a number of cancertypes especially hematological tumors
