Osteoarthritis (OA) is a chronic inflammatory disease that causes dysfunction of cartilage and joints. Activation of the immune system and the development of inflammatory responses are among the main pathogenesis of the disease. It has been proved that the expression of pro-inflammatory cytokine in patients is increased and leads to the destruction of cartilage in the joints of patients. Therefore, the aim of this study was to evaluate the effect of menthol-silver nanoparticles۱ on gene expression of TNF-a۲, iL-۱β۳, COX-۲۴, and iNO۵ in bovine fibroblast-like synoviocytes۶ and production of PGE۲۷ and NO۸ in THP-۱. The usage of nanotechnology in medicine particularly for drug delivery is revealed to have numerous benefits. It has been reported to have anti-inflammatory, diuretic, and antihypertensive activity. Plant mediated biological synthesis of nanoparticles is simple and eco-friendly method. BFLSc and THP-۱ cells were first incubated with LPS to induce inflammation. Then, the expression of il۱-β, il-۶, iNOS, TNF-α and COX-۲ and production of PGE۲ and No were evaluated using RT-PCR and ELISA respectively. This work therefore was aimed to synthesize Menthol-Ag-NPs, and evaluate its anti-inflammatory activity. Menthol- Ag-NPs۹ were synthesized by short term (۱ day) interaction of Menthol (۱ ml) with ۲mM AgNO۳ solution and centrifuged to obtain silver nanoparticles. The nanoparticles were characterized by UV-Visible spectrophotometer, FTIR and Scanning Electron Microscopy. Further, the Menthol-AgNPs were evaluated for anti-inflammatory activity by ex-vivo method. The results showed Menthol-AgNPs downregulated TNF-α, iL-۱β, COX-۲, and iNOS expression in synoviocytes this suppression was paralleled by a significant reduction in PGE۲ and NO, respectively, in the cellular supernatant of THP-۱.
