Effects of Punicic acid on expression of Cox-۱ in KATO III cells and Cox-۲ in Bovine Fibroblast-like Synoviocyte

Abstract:

Gastric ulcer is a lesion that may develop in gastric mucosa and submucosa or duodenum. Gastric ulcers develop because of increased acid secretion due to alcohol abuse, nonsteroidal anti-inflammatory drugs (NSAIDs)۱ such as sedatives and aspirin, continuous and prolonged hunger, continuous stress, infection with Helicobacter pylori, and poor diet. Today, drugs such as glucocorticoids (GCs) and NSIADs they are worldwide prescribed drugs for many musculoskeletal conditions, such as osteoarthritis and inflammatory rheumatic diseases, but have serious and life-threatening side effects. The aim of the current study is to evaluate the effects of punicic acid on COX-۱, COX-۲ in KATO III and bovine fibroblast-like Synoviocytes cells (BFLSc)۲, and to compare it with glucocorticoids and ibuprofen. Synovial cells were isolated from synovial fluid of the radiocarpal joint cartilage of an ۸-month-old Holstein cow. KATO III cells were purchased from Pasteur Institute of Iran. Cells were cultivated and exposed to lipopolysaccharide [LPS]۳ stimulation without, or in the presence of, dexamethasone, ibuprofen and punicic acid. The gene expressions of COX-۱۴, COX-۲۵ were evaluated by RT- PCR۶. The results of this study showed that LPS-stimulated BFLSc and treated with puicicic acid cells reduce COX-۲ gene expression. On the other hand, KATO III cells treated with punicic acid and stimulated with LPS did not increase COX-۱gene expression. The expression level of COX-۲ in LPS-stimulated cells treated with punicic acid showed a significant decrease compared to dexamethasone and ibuprofen. Panic acid shows the prevention of increased production and expression of COX-۱ in KATO III cells and on the other hand in stimulated cells does not show a change in reduced production and expression of COX-۱ gene.
