Lactobacillus is the largest group of lactic acid bacteria and has many applications in industry and health. Soil rhizosphere bacteria play a role in hormone production, dissolution of plant food compounds, facilitation of nutrient absorption, and reduction of plant diseases. The isolation of Lactobacilli from dairy and non-dairy sources has been reported. Soil is one of the important sources rich in all kinds of microorganisms. Studies show that soil rhizosphere bacteria, including Lactobacilli, are effective in plant growth and health.This study aims to investigate the biochemical analysis of Lactobacilli isolated from the rhizosphere of grapevine by preparing ۰ to ۱۰-۷ dilutions of the sample and transferring it to MRS medium (Lactobacillus specific culture medium) by phenotypic methods (gram staining and biochemical tests such as Catalase, Oxidase, KOH, SIM, TSI, Simmons’ citrate, MR-VP, and fermentation of carbohydrates). Among the ۲۰ isolated strains, ۱۲ strains were gram-positive, bacilli-shaped, Oxidase, and catalase negative. This research showed that the rhizosphere of grapevine soil is a suitable source for the identification of Lactobacillus bacteria.
