Abstract:

Medicinal plants have been used since long for mankind against various infectious and noninfectiousdiseases because they contain natural bioactive components for therapeutic value.Aim of this research is bioactivity determination, MIC and MBC extracts obtained from theleaves of Artemisia annua Linn. against standard strains of bacteria andclinical isolates.Theleaves (40 g) A. annua were dried at 25°C in the shade and extracted by using Soxhlet systemThe extract was weighed and stored at 4 – 6°C in dark until use. Antibacterial activityscreening for crude extract was screened against 8 bacteria using Mueller-Hinton agar plates(MHA, oxoid) (for all the bacteria) previously inoculated with 18 hold Nutrient broth (NB,oxoid) culture (0.5 McFarland Standard) for the bacteria.To identify the phytochemicalderivatives in the extract, standard phytochemical screening was performed. MIC assay of theextracts were also determined with using the same method except that the paper discs weresoaked in different concentrations of crude extract dispersed in water (10 - 2000 μL). Afterincubating at 24 h at 37°C, the MIC of each sample was determined by measuring the opticaldensity in the spectrophotometer (620 nm), and comparing the result with those of the noninoculated MHB (oxoid 2009). For determination of MBC, 1 ml was pipetted from themixture obtained in the determination of MIC tubes which did not show any growth andstreaked on MHA and incubated for 24 h . The antibacterial activity of A. annua leaf extractswere examined against tested microorganisms. In this work, according to results, the extract50 mg/mL was indicated more activity in comparison with Chloramphenicol against clinicalisolates of S. aureus. Extract 50 mg/mL showed the highest activity against B.cereuscompared with the antibiotics tested. Extract (50 mg/mL ) was found to have some activity asthat of the P.aeruginosa.in our tests; penicillin showed any activity against clinical isolates ofS. aureus, UPEC, P.aeruginosa and also standard strains of S.aureus and E.coli. Extract andstreptomycin have been shown similar activity at equal concentrations against Bacillus sp.The large size of the zone of inhibition has indicated that streptomycin have more potencycompared with extract 50 mg/mL against P. aeruginosa.
